用ISSR-PCR(Inter Simple Sequence Repeats PCR)技术对栉孔扇贝Chlamys farreri♀和虾夷扇贝Pa-tinopecten yessoensis♂及其单对杂交子一代担轮幼虫进行了分析。结果表明:从20个ISSR引物中筛选出4个引物对栉孔扇贝♀、虾夷扇贝♂及其单对杂交子一代担轮幼虫进行PCR扩增,共得到153个清晰稳定的扩增位点,其中母本栉孔扇贝的特异位点有38个,父本虾夷扇贝的特异位点有32个。对ISSR扩增图谱的分析结果证实,所研究的杂交子代是栉孔扇贝和虾夷扇贝的杂交种。杂交子代与母本栉孔扇贝的平均遗传相似性系数为0.6086,与父本虾夷扇贝的平均遗传相似性系数为0.4402,表明杂交子代在遗传上偏向于母本栉孔扇贝。
The larvae of Chlamys farreri♀ and Pa-tinopecten yessoensis♂ and their single pairs of hybrids were detected by ISSR-PCR (Inter Simple Sequence Repeats PCR). The results showed that 4 primers were screened from 20 ISSR primers for PCR amplification of Zhikong Scallop, Scallop ♂ and their single-hybrid hybrid generation, and 153 clear and stable amplification loci were obtained , Of which 38 were specific for the mother Chlamys farreri and 32 were specific for the male parent Scallop. Analysis of the ISSR amplification pattern confirmed that the hybrid progeny studied was a cross between Chlamys farreri and Scallop shrimp. The average genetic similarity coefficient between hybrid offspring and the parent Chlamys farreri was 0.6086, and the average genetic similarity coefficient with the male parent was 0.4402, indicating that the hybrid offspring were genetically biased towards the mother Chlamys farreri.
