目的比较转人乳铁蛋白基因(hLf)大米与其亲本大米在营养成分上的差异,并观察其是否具亚慢性毒性作用。方法对大米中的主要成分及矿物质、维生素进行测定并比较。将初断乳Wistar大鼠按性别、体重随机分为4组饲喂90天,自由进食与饮水。依据大米成分分析结果配制饲料:转hLf大米高剂量饲料(A组,大米添加量为71.45%)、转hLf大米中剂量饲料(B组,大米添加量为35.725%)、亲本大米高剂量饲料(C组,大米添加量为71.01%)和AIN-93G对照饲料(D组)。观察大鼠的体重、进食量、血常规、血生化、脏器系数、骨密度情况,并对大鼠的脏器进行病理学检查。结果转hLf大米与亲本大米在总的主要营养成分、矿物质、维生素上没有显著差异,但在乳铁蛋白及Fe含量上具明显差异。转hLf大米组血常规(WBC、HGB、RBC、MCH)、血生化(AST、Glu)、脏器系数、骨密度的个别指标与亲本对照组或AIN-93对照组存在显著差异(P<0.05);其他观察指标与两个对照组均无显著差异。结论部分指标虽然存在组间差异,但仍在正常范围内,未发现转hLf大米对健康具有毒副作用。
Objective To compare the difference in nutritional content of rice transformed with lactoferrin gene (hLf) and its parent rice and observe whether it has subchronic toxicity. Methods The main components of rice and minerals, vitamins were measured and compared. The weaned Wistar rats were randomly divided into 4 groups according to their sex and weight for 90 days and were fed with water freely. According to the result of rice component analysis, the feed was prepared: hLf rice high-dose feed (A group, 71.45% rice added), medium hLf rice feed (B group, 35.725% rice added) C group, the amount of rice added was 71.01%) and AIN-93G control group (D group). The body weight, food intake, blood routine, blood biochemistry, organ coefficient and bone mineral density of the rats were observed. Pathological examination of the organs was performed. Results There was no significant difference in total main nutrients, minerals and vitamins between hLf rice and parental rice, but there was a significant difference in lactoferrin and Fe content. There were significant differences in individual indexes of blood routine (WBC, HGB, RBC, MCH), blood biochemistry (AST, Glu), organ coefficient and bone mineral density in hLf rice group compared with parental control group or AIN-93 control group ); Other indicators and no significant difference between the two control groups. Conclusion Although there are some differences between groups, some indicators are still in the normal range, and no hLf rice has been found to have toxic and side effects on health.
