目的:探讨伤科接骨片在下颌骨缺损修复中的作用机制。方法:健康雄性新西兰兔72只,随机分为正常对照组(24只)、模型对照组(24只)和伤科接骨片组(24只),建立下颌骨缺损模型。正常对照组、模型对照组动物给予正常饲料,伤科接骨片组动物给予伤科接骨片饲料。分别于建模后7、14、28、56天处死相同例数动物(n=6),收集下颌骨缺损处骨组织块。采用免疫组织化学技术检测骨保护素(Osteoprotegerin,OPG)、骨保护素配体(Osteoprotegerin ligand,OPGL)表达变化。结果:与正常对照组比较,模型对照组OPG、OPGL表达分别于建模后第7、14天显著提高(P<0.05),伤科接骨片组OPG表达于建模后第7、14、28、56天均显著提高(P<0.05),OPGL表达则提高不明显(P>0.05);与模型对照组比较,伤科接骨片组OPG表达于建模后第14、28、56天均显著提高(P<0.05),OPGL表达于建模后第14天显著降低(P<0.05),OPG/OPGL比值在建模后第14、28、56天显著上升(P<0.05)。结论:伤科接骨片可能通过影响OPG、OPGL的表达而促进下颌骨缺损修复。
Objective: To explore the mechanism of Shangke Jiegu tablets in the repair of mandibular defects. Methods: A total of 72 healthy male New Zealand rabbits were randomly divided into normal control group (24 rats), model control group (24 rats) and Shangke Jiegu tablets group (24 rats). The normal control group and the model control group were given the normal diet, and the animals in Shangkejiegu tablet group were given Shangkejiegu tablets. The same number of animals (n = 6) were sacrificed at 7, 14, 28, and 56 days after modeling, and the bone tissue pieces of the mandible defect were collected. Immunohistochemistry was used to detect the expression changes of osteoprotegerin (Osteoprotegerin, OPG) and Osteoprotegerin ligand (OPGL). Results: Compared with the normal control group, OPG and OPGL expression in the model control group were significantly increased (P <0.05) on the 7th and 14th day after modeling, respectively. OPG expression in Shangkejiegu tablet group was significantly higher than that in the control group after 7, 14, 28 (P <0.05), while the expression of OPGL did not increase significantly (P> 0.05). Compared with the model control group, OPG expression in Shangke Jiegu tablet group was significantly higher than that of the control group on the 14th, 28th and 56th day (P <0.05). The expression of OPGL decreased significantly on the 14th day after modeling (P <0.05). The ratio of OPG / OPGL increased significantly on the 14th, 28th and 56th day after modeling (P <0.05). Conclusion: Shangke Jiege tablet may promote the repair of mandibular defects by affecting the expression of OPG and OPGL.
