为探讨ATRA和IFNα合用对体外慢性粒细胞白血病(CML)白血病细胞增殖的影响,作者应用ATRA、IFNα或者两药联合对初诊Ph(+)CML慢性期患者的CD34~+细胞进行了深入的体外研究。 材料和方法 血细胞取自16名未经治疗的初诊CML慢性期患者,其Ph(+)中期分裂相均100%。先用Ficoll-hypaque密度梯度离心分离单个核细胞,继之用免疫磁性分离法收集CD34~+细胞,其纯度大于90%。
To investigate the effect of combination of ATRA and IFNα on the proliferation of chronic myelogenous leukemia (CML) leukemia cells in vitro, the authors used ATRA, IFNα, or a combination of two drugs to perform in-depth in vitro CD34~+ cells in patients with newly diagnosed Ph(+) CML chronic phase. the study. Materials and Methods Blood cells were obtained from 16 untreated, initially diagnosed chronic CML patients with 100% Ph(+) metaphase. The mononuclear cells were first separated by Ficoll-hypaque density gradient centrifugation, followed by immunomagnetic separation to collect CD34~+ cells with a purity greater than 90%.
