目的探讨止颤胶囊对帕金森病(PD)模型大鼠脑内单胺氧化酶B(MAO-B)表达的调节作用。方法应用脑内定位注射6-羟基多巴胺(6-OHDA)方法制备部分损伤PD模型;采用逆转录聚合酶链反应(RT-PCR)对PD大鼠脑内MAO-B mRNA的表达进行分析;采用生化法对脑内MAO-B活性进行测定。结果从给药后第10天开始,模型组MAO-B mRNA表达明显增加,扩增产物的量为止颤胶囊的4.728倍,远远高于止颤胶囊组和对照组;MAO-B的活性检测结果与RT-PCR分析结果一致,止颤胶囊对大鼠脑内MAO-B活性抑制率大于40%。结论止颤胶囊对PD大鼠脑内MAO-B mRNA表达和活性有下调作用,可使6-OHDA引起的脑内MAO-B mR-NA高表达得到抑制。
Objective To investigate the effect of Zhituo Capsule on the expression of monoamine oxidase B (MAO-B) in the brain of Parkinson’s disease (PD) model rats. Methods PD model was induced by injection of 6-hydroxydopamine (6-OHDA) intracerebrally. The expression of MAO-B mRNA in brain of PD rats was analyzed by reverse transcription-polymerase chain reaction (RT-PCR) Biochemical assay of MAO-B activity in the brain. Results From the 10th day after administration, the MAO-B mRNA expression in the model group increased significantly, and the amount of the amplified product was 4.728 times that of the Zhifu Capsule group and the control group. The activity of MAO-B Results The results of RT-PCR analysis showed that Zhiru capsule inhibited MAO-B activity in rat brain more than 40%. Conclusion Zhigan capsule can down-regulate the expression and activity of MAO-B mRNA in the brain of PD rats, and inhibit the expression of MAO-B mR-NA induced by 6-OHDA in the brain.
