目的观察α-硫辛酸对糖尿病大鼠腺苷酸活化蛋白激酶(AMPK)的影响,探讨其改善糖尿病代谢异常的作用。方法将大鼠分为正常对照(NC)组、T2DM(T2DM)组、糖尿病α-硫辛酸干预(LA)组。Western blot检测下丘脑及骨骼肌组织AMPKα及ph-AMPKα蛋白的表达,RT-PCR测AMPKαmRNA表达。结果与NC组比较,T2DM组骨骼肌、下丘脑内AMPK表达水平(21%和22%)及活性(37%和21%)降低(P<0.05),血糖、血脂水平升高;与T2DM组比较,LA组骨骼肌内AMPK表达水平(20%)及活性(49%)升高(P<0.05),而下丘脑内AMPK表达水平(17%)及活性(36%)降低(P<0.05),血糖、血脂水平降低。结论α-硫辛酸可激活骨骼肌AMPK,抑制下丘脑AMPK活性,可能是其改善糖脂代谢异常的部分机制。
Objective To investigate the effect of α-lipoic acid on adenylate-activated protein kinase (AMPK) in diabetic rats and to explore its effect on improving the abnormal metabolism of diabetes mellitus. Methods The rats were divided into normal control (NC) group, T2DM (T2DM) group and diabetic α-lipoic acid intervention (LA) group. Western blot was used to detect the expression of AMPKα and ph-AMPKα in hypothalamus and skeletal muscle, and the expression of AMPKα mRNA was detected by RT-PCR. Results Compared with NC group, the levels of AMPK (21% and 22%) and activity (37% and 21%) in skeletal muscle and hypothalamus of T2DM group were significantly lower (P <0.05) Compared with the control group, the AMPK expression level (20%) and activity (49%) were increased in the skeletal muscle of the LA group (P <0.05), while the AMPK expression level (17%) and activity ), Blood glucose, blood lipid levels decreased. CONCLUSION: Alpha-lipoic acid can activate AMPK in the skeletal muscle and inhibit AMPK activity in the hypothalamus, which may be part of the mechanism of improving the abnormal glucose and lipid metabolism.
