‘青砧一号’是杂交育成的苹果矮化砧木新品系。本文探讨了影响‘青砧一号’离体再生的条件,建立了高效离体再生体系。结果表明,以叶片为外植体,远轴面接触培养基,最佳不定芽诱导分化培养基:MS+5.0 mg·L-16-BA+0.3 mg·L-1NAA;最佳继代增殖培养基:MS+1.0 mg·L-16-BA+0.1 mg·L-1IAA+20 g·L-1蔗糖,不定芽的增殖倍数为2.7;最佳生根培养基:1/2MS+1.0 mg·L-1IAA+20 g·L-1蔗糖,生根率达100%,平均生根数达5.7条。炼苗后移入珍珠岩、蛭石、草炭等体积混合的基质中,20 d后移栽成活率达60%。
’Aventurine’ is a new breed of apple dwarfing rootstock hybridized. In this paper, the conditions affecting the in vitro regeneration of ’Aventurine One’ were discussed, and an efficient regeneration system in vitro was established. The results showed that the optimal medium for adventitious bud differentiation was MS + 5.0 mg · L-16-BA + 0.3 mg · L-1 NAA with the leaves as the explants and the distal surface contacting the medium. The best subculture The multiplication rate of adventitious buds was 2.7. The best rooting medium was 1 / 2MS + 1.0 mg · L -1 -1 IAA + 20 g · L -1 sucrose, rooting rate of 100%, the average number of roots 5.7. In the medium of perlite, vermiculite, peat and other mixed volume, the survival rate after transplanting was 60% after 20 days.
