目的 :检测血管内皮生长因子C(vascularendothelialgrowthfactorC ,VEGF C)mRNA和蛋白在人乳腺癌细胞株MCF 7及其耐药株MCF 7/Adr中的定位、定性表达。方法 :根据VEGF C基因序列 ,设计合成地高辛标记的特异性寡核苷酸探针 ,运用原位杂交方法检测培养的细胞株MCF 7和MCF 7/Adr中VEGF CmRNA的表达 ;并运用免疫组织化学方法检测了两种细胞中VEGF C蛋白的表达。结果 :原位杂交检测到MCF 7和MCF 7/Adr细胞的胞浆中有阳性蓝色颗粒 ,免疫组化检测发现两种细胞的胞浆中均有阳性棕黄色颗粒 ,而阴性对照细胞的胞浆中则均无阳性颗粒。结论 :人乳腺癌细胞株MCF 7及其耐药株MCF 7/Adr细胞能够转录VEGF CmRNA并在其细胞浆中翻译合成相应的蛋白。
OBJECTIVE: To detect the localization and expression of vascular endothelial growth factor C (VEGF C) mRNA and protein in human breast cancer cell line MCF-7 and its multi-drug resistance strain MCF 7 / Adr. Methods: According to the sequence of VEGF C gene, a synthetic digoxigenin-labeled oligonucleotide probe was designed. The expression of VEGF mRNA in cultured MCF 7 and MCF 7 / Adr cells was detected by in situ hybridization. Histochemical methods were used to detect VEGF C protein expression in both cells. Results: The positive blue particles were detected in the cytoplasm of MCF 7 and MCF 7 / Adr cells by in situ hybridization. Immunocytochemistry showed that there were positive brown granules in the cytoplasm of both MCF 7 and MCF 7 / Adr cells, while the cells of negative control cells No positive particles in the pulp. Conclusion: The human breast cancer cell line MCF 7 and its multidrug-resistant MCF 7 / Adr cells can transcribe VEGF mRNA and synthesize their corresponding proteins in their cytoplasm.
