目的对粤蛇葡萄Ampelopsis cantoniensis地上部分的化学成分及其抗血管生成活性进行研究。方法采用多种柱色谱技术进行分离纯化,通过波谱分析鉴定化合物结构,并用斑马鱼模型筛选其抗血管生成活性。结果从粤蛇葡萄地上部分90%乙醇提取物的醋酸乙酯萃取部分中分离得到15个化合物,分别鉴定为粤蛇葡萄醇(1)、圆柚酮(2)、aromadendrane-4β,10β-diol(3)、脱落酸(4)、12-oxo-hardwickiic acid(5)、白桦脂酸(6)、platanic acid(7)、香草酸(8)、白藜芦醇(9)、nectandrin B(10)、nectandrin A(11)、3,5,7-三羟基色原酮(12)、5,7,3′,4′,5′-五羟基二氢黄酮(13)、花旗松素(14)、杨梅苷(15)。结论化合物1为新的桉烷倍半萜,命名为粤蛇葡萄醇;化合物2~7、10~12为首次从蛇葡萄属中分离得到;其余化合物为首次从该植物中分离得到。化合物9、11、12表现出一定的抗斑马鱼血管生成活性。
OBJECTIVE To study the chemical constituents and anti-angiogenic activities of aerial parts of Ampelopsis cantoniensis in Guangdong snake grape. Methods A variety of column chromatography was used to separate and purify the compounds. Compounds were identified by spectral analysis and their antiangiogenic activity was screened by zebrafish model. Results Fifteen compounds were isolated from the ethyl acetate extracts of 90% ethanolic extracts of Yue snake grape, and identified as Yue snake grape wine (1), melon ketone (2), aromadendrane-4β, 10β-diol (4), 12-oxo-hardwickiic acid (5), betulinic acid (6), platanic acid (7), vanillic acid (8), resveratrol (9), nectandrin B 10), nectandrin A (11), 3,5,7-trihydroxychromone (12), 5,7,3 ’, 4’, 5’-pentahydroxyflavone (13) 14), myricitrin (15). CONCLUSION Compound 1 is a new eucalyptol sesquiterpenoid, named Cantharelol; Compounds 2 ~ 7,10 ~ 12 were isolated from the genus Snake for the first time; the rest of the compounds were isolated from this plant for the first time. Compounds 9, 11, 12 showed some anti-zebrafish angiogenic activity.
