目的探讨低剂量硝酸镧对糖尿病(diabetes mellitus,DM)大鼠血清中过氧化物酶体增殖物激活受体-γ(peroxisome proliferator-activated receptor-gamma,PPAR-γ)和葡萄糖转运蛋白4(glucose transporter protein 4,GLUT4)蛋白表达水平的影响。方法将40只健康清洁级Wistar雄性大鼠随机分为3组,分别为对照组(10只)、糖尿病组(18只)和硝酸镧组(12只)。糖尿病组和硝酸镧组腹腔注射50 mg/kg链脲佐菌素制备糖尿病模型;硝酸镧组采用灌胃方式染毒0.2mg/kg硝酸镧,对照组及糖尿病组均每日给予等剂量的生理盐水,每天1次,连续染毒1个月。采用流式细胞学检测大鼠全血中PPAR-γ和GLUT4蛋白的阳性百分率;采用ELISA法检测大鼠血清中PPAR-γ和GLUT4蛋白的表达水平。结果与对照组比较,糖尿病组大鼠全血中PPAR-γ和GLUT4蛋白阳性百分率及血清中PPAR-γ和GLUT4蛋白表达水平均降低,差异有统计学意义(P<0.01);但硝酸镧组大鼠全血中PPAR-γ和GLUT4蛋白阳性百分率及血清中PPAR-γ和GLUT4蛋白表达水平均无明显变化。硝酸镧组大鼠全血中PPAR-γ和GLUT4蛋白阳性百分率及血清中PPAR-γ和GLUT4蛋白表达水平均高于糖尿病组,差异有统计学意义(P<0.01)。结论低剂量(0.2 mg/kg)硝酸镧可通过促进糖尿病大鼠全血及血清中PPAR-γ和GLUT4蛋白的表达调节大鼠糖代谢。
Objective To investigate the effects of low dosage of lanthanum nitrate on the serum levels of peroxisome proliferator-activated receptor-gamma (PPAR-γ) and glucose 4 (glucose) in diabetic mellitus (DM) transporter protein 4, GLUT4) protein expression. Methods Forty healthy male Wistar rats were randomly divided into three groups: control group (n = 10), diabetes group (n = 18) and lanthanum nitrate group (n = 12). Diabetic group and lanthanum nitrate group were given intraperitoneal injection of streptozotocin 50 mg / kg to prepare diabetic model; lanthanum nitrate group was intragastrically administered 0.2 mg / kg lanthanum nitrate, the control group and diabetic group were given equal daily doses of physiology Saline, 1 day, continuous exposure to 1 month. The percentages of PPAR-γ and GLUT4 in whole blood of rats were detected by flow cytometry. The expression of PPAR-γ and GLUT4 protein in serum was detected by ELISA. Results Compared with the control group, the percentages of PPAR-γ and GLUT4 in whole blood and the levels of PPAR-γ and GLUT4 in the whole blood of diabetic rats decreased significantly (P <0.01) The percentages of PPAR-γ and GLUT4 in whole blood of rats and the levels of PPAR-γ and GLUT4 in serum did not change significantly. The positive percentage of PPAR-γ and GLUT4 protein in whole blood and the levels of PPAR-γ and GLUT4 in serum of the Lanthanum nitrate group were significantly higher than those of the diabetic group (P <0.01). Conclusion Low dose (0.2 mg / kg) Lanthanum nitrate can regulate glucose metabolism in rats by promoting the expression of PPAR-γ and GLUT4 protein in whole blood and serum of diabetic rats.
