用抗弓形虫单克隆抗体Dot-ELISA检测350例孕妇和18例娩出畸形儿的产妇血清中的弓形虫循环抗原(CAg),同时用Dot-ELISA检测这批血清中的弓形虫抗体IgG。350例孕妇血清中弓形虫CAg阳性3例,阳性率为086%。弓形虫IgG阳性18例,阳性率为514%。332份弓形虫IgG阴性血清CAg均为阴性。18例IgG阳性血清中3例CAg阳性,抗体滴度分别为1∶40,1∶40,1∶160,提示孕妇有早期活动性感染;还提示检测弓形虫IgG阳性抗体滴度不高时应作CAg检测,不可轻易作为既往感染以免漏诊了孕妇早期感染致使胎儿感染弓形虫。18例产妇血清中有2例血清CAg和IgG均为阳性,并分别从畸胎组织和脐血和羊水中分离出弓形虫。5例疟疾病人血清均无交叉反应。实验结果显示抗弓形虫单克隆抗体Dot-ELISA检测弓形虫CAg敏感性高、特异性强、方法简易、快速,是弓形虫病早期临床诊断的较理想方法。
Toxoplasma gondii circulation antigen (CAg) was detected in 350 pregnant women and 18 maternal women with deformed fetuses by Dot-ELISA using Dot-ELISA. Dot-ELISA was used to detect Toxoplasma IgG in these sera. Three cases of Toxoplasma gondii CAg were detected in 350 pregnant women, the positive rate was 086%. Toxoplasma gondii IgG positive in 18 cases, the positive rate was 5 14%. 332 Toxoplasma gondii IgG negative serum CAg were negative. 18 cases of IgG-positive serum in 3 cases of CAg-positive, antibody titers were 1:40,1:40,1:160, suggesting that pregnant women have early active infection; also suggested that Toxoplasma gondii IgG positive antibody titers should not be high For CAg test, can not easily as a past infection in order to avoid misdiagnosis of early pregnancy caused by fetuses infected with Toxoplasma gondii. Two of 18 maternal serums were positive for serum CAg and IgG, and Toxoplasma gondii were isolated from teratocytes, cord blood and amniotic fluid respectively. No serum was detected in 5 malaria patients. The experimental results showed that the detection of Toxoplasma gondii CAg by Dot-ELISA against Toxoplasma gondii is highly sensitive and specific, and the method is simple and rapid. It is an ideal method for the early clinical diagnosis of toxoplasmosis.
