目的用不同剂量的141株鼠疫菌对实验豚鼠鼠蹊部皮下接种后引起的病理特征变化,并进行其组织形态学观察。方法采用141株鼠疫菌分25个(低剂量组)、100个(中剂量组)、5 000个(高剂量组)三个剂量组接种成体实验豚鼠鼠蹊部皮下,接种后,有病死的立即取材、未病死的14 d分别处死实验豚鼠取材。取肝脏、脾脏、肺脏、肾脏及心脏等实质性脏器固定灭菌做病理切片,HE染色,光镜下观察,同时对所取的脏器材料做鼠疫细菌学培养和鼠疫噬菌体裂解实验,以确定各脏器是否含菌,豚鼠是否是特异性死亡。结果接种不同剂量141株强毒鼠疫菌的实验豚鼠的肝脏、脾脏、肺脏、肾脏和心脏均分离出鼠疫菌;其组织巨检均有显著的病理变化,镜检除心脏外均有明显的病理变化,且呈不同程度的充血、出血、脂肪变性及组织坏死表现。结论 141株鼠疫菌的不同剂量组均引起实验豚鼠急性炎症反应,豚鼠肝、脾、肺、肾、心等实质性脏器的病理变化显著,并随接种剂量增大而出血及组织坏死逐渐严重。
Objective To investigate the pathological changes of 141 strains of Yersinia pestis caused by subcutaneously inoculation of experimental guinea pigs and to observe their histomorphology. Conclusions The 141 strains of Yersinia pestis at different dosage groups all induced acute inflammatory reaction in guinea pigs. The pathological changes of liver, spleen, lung, kidney, heart and other organs in guinea pigs were significant, and the bleeding and tissue necrosis gradually increased with the increase of inoculation dose .
