目的 为了寻找一种新的高效制备高特异性高亲和力单克隆抗体的工具 ,提高工作效率 ,增强诊断试剂科学性和准确性。方法 利用无菌隔离器技术 ,无抗原饲料 (由氨基酸、碳水化合物、维生素、必需矿物质、植物油等组成的分子量小于 80 0 0的化合物 )喂养无菌小鼠 ,将无菌小鼠培育成无抗原小鼠。结果 经ELISA方法检测该小鼠血清中IgG的含量 ,其含量是CV和GF小鼠的 5 % - 10 %。结论 无抗原技术已完全成熟 ,无抗原小鼠培育成功。由于该无抗原小鼠未接受过外源性抗原的刺激 ,可利用该动物制备高特异性、高亲和力的单克隆抗体
Objective To find a new tool to efficiently prepare monoclonal antibodies with high specificity and high affinity, improve work efficiency and enhance the scientificity and accuracy of diagnostic reagents. Methods Sterile mice were aseptically challenged with sterile antigen-free diet (compounds with a molecular weight of less than 80,000 consisting of amino acids, carbohydrates, vitamins, essential minerals, vegetable oils, etc.) Antigen mouse. Results The content of IgG in the serum of mice was detected by ELISA, which was 5% -10% of the CV and GF mice. Conclusion No antigen technology has been fully mature, no antigen in mice successfully.
