目的:探讨酵母RNA的肝脏保护、免疫调节以及延缓衰老的作用。方法:①小鼠四氯化碳腹腔注射,建立急性肝损伤模型,行ALT、AST检测及肝组织学检查;②用5-氟尿嘧啶建立免疫抑制模型,行巨噬细胞吞噬率、血清TNF-α、IL-2检测,计算脾脏指数、胸腺指数;③30只22月龄Wister大鼠随机分为正常组、酵母RNA组和VitE组,检测脑匀浆的SOD、MDA。结果:酵母RNA组小鼠血清ALT、AST明显低于模型组(P<0.01~P<0.05),肝组织中炎症细胞浸润、肝细胞坏死和变性损害也显著轻于模型组;与5-氟尿嘧啶组相比,酵母RNA可显著提高巨噬细胞吞噬率、脾脏指数、胸腺指数(P<0.01)、血清TNF-α和IL-2(P<0.05);和正常组相比,酵母RNA可显著提高老年大鼠脑匀浆的SOD,降低MDA(P<0.01)。结论:酵母RNA对急性肝损伤有一定保护作用,对免疫低下小鼠有免疫增强作用,对老年大鼠有延缓衰老的作用。
OBJECTIVE: To investigate the liver protection, immunomodulatory and anti-aging effects of yeast RNA. METHODS: ①Methods: ①Acute liver injury model was induced by intraperitoneal injection of carbon tetrachloride in mice. ALT, AST and liver histological examination were performed. ②The immunosuppressive model was established by 5-fluorouracil, and the phagocytosis rate of macrophages, serum TNF-α , Spleen index and thymus index were calculated by IL-2 test. (3) Thirty Wistar rats of 22 months old were randomly divided into normal group, yeast RNA group and VitE group. Results: Serum ALT and AST levels in yeast RNA group were significantly lower than those in model group (P <0.01 ~ P <0.05). Inflammatory cell infiltration and hepatocyte necrosis and degeneration in liver tissue were also significantly lower than those in model group. Yeast RNA significantly increased macrophage phagocytosis, spleen index, thymus index (P <0.01), serum TNF-α and IL-2 (P <0.05) compared with the control group Increased SOD in brain homogenates of aged rats and decreased MDA (P <0.01). Conclusion: Yeast RNA has a protective effect on acute liver injury, immune enhancement in immunocompromised mice, and anti-aging effects on aged rats.
