采用多聚酶链反应(PCR)技术检测水中肠道病毒前,肠道病毒应先行浓集。本研究成功地用金葡菌固相抗体对水中肠道病毒的指标菌f_2噬菌体进行浓集。(1)金葡菌固相抗体先行混悬于水样中,与f_2噬菌体结合,然后在3000r/min下离心10min。1ml金葡菌固相抗体约可吸附3000PFU以上的f_2噬菌体。(2)蒸馏水、NaCl溶液、磷酸盐缓冲液(PBS)及三倍肉汤对金葡菌固相抗体吸附f_2的效果均无明显影响,吸附率均在93.7%以上。(3)用PCR技术对金葡菌固相抗体捕获的脊髓灰质炎病毒I型疫苗株(PVI)亦可检测。
Before using polymerase chain reaction (PCR) to detect waterborne enterovirus, enterovirus should be concentrated first. In this study, Staphylococcus aureus solid-phase antibody was successfully used to collect the indicator phage f_2 phage of enterovirus in water. (1) Staphylococcus aureus solid-phase antibody was first suspended in water and bound to f 2 bacteriophage, then centrifuged at 3000 r / min for 10 min. 1ml Staphylococcus aureus solid phase antibody can adsorb more than 3000PFU phage f_2. (2) The effects of distilled water, NaCl solution, phosphate buffered saline (PBS) and triple broth on the adsorption of f 2 by Staphylococcus aureus solid-phase antibody had no obvious effect. The adsorption rates were above 93.7%. (3) The poliovirus type I vaccine strain (PVI) captured by Staphylococcus aureus solid phase antibody by PCR can also be detected.
