目的研究全反式维甲酸(ATRA)对肺腺癌A549细胞凋亡的影响及机制。方法MTT法观察ATRA对肺癌细胞株A549的生长抑制作用,流式细胞仪观察ATRA诱导A549细胞凋亡作用和对细胞周期的影响,Western Blot分析凋亡相关蛋白表达。结果ATRA抑制A549的增殖呈剂量依赖性;ATRA可诱导A549细胞凋亡,凋亡过程伴随caspase-3、caspase-9表达增加,而Bcl-2、Bcl-Xs/LP的表达无明显变化。结论ATRA可明显抑制A549细胞的增殖,可通过上调caspase-9、caspase-3的表达促进A549细胞凋亡。
Objective To investigate the effect of all-trans retinoic acid (ATRA) on the apoptosis of lung adenocarcinoma A549 cells and its mechanism. Methods The inhibitory effect of ATRA on the growth of lung cancer cell line A549 was observed by MTT assay. The effect of ATRA on A549 cell apoptosis and the cell cycle were observed by flow cytometry. The expression of apoptosis related protein was analyzed by Western Blot. Results ATRA inhibited the proliferation of A549 in a dose-dependent manner. ATRA could induce the apoptosis of A549 cells. The apoptosis accompanied with the increase of caspase-3 and caspase-9, while the expression of Bcl-2 and Bcl-Xs / LP did not change significantly. Conclusion ATRA can significantly inhibit the proliferation of A549 cells and promote the apoptosis of A549 cells by up-regulating the expression of caspase-9 and caspase-3.
