目的 探讨体外照射后造血细胞表面IL 1 1受体 (IL 1 1R)蛋白和基因表达水平的变化 ,为临床更合理有效地应用rhIL 1 1治疗急性放射病提供理论和实验依据。方法 采用流式细胞术及RT PCR法检测了不同照射剂量和照射后不同时间K5 6 2细胞表面IL 1 1R蛋白和基因表达水平的变化。结果 随着照射剂量的升高 ,K5 6 2细胞表面IL 1 1Rα蛋白的表达量逐渐升高 ,其中 5Gy和 1 0Gy照射组照射后 2 4hIL 1 1Rα蛋白表达水平较正常显著增高。结论 照射后 2 4hIL 1 1Rα蛋白及基因表达水平升高可能是机体自我保护的一种适应性反应 ,为早期应用IL 1 1提供了物质基础
Objective To investigate the changes of IL-1 receptor (IL-1R) protein and gene expression on hematopoietic cells after in vitro irradiation, so as to provide theoretical and experimental basis for the clinical application of rhIL-1 in the treatment of acute radiation sickness. Methods The changes of IL-1R protein and gene expression on K562 cells were detected by flow cytometry and RT-PCR at different doses and time. Results With the increase of irradiation dose, the expression of IL-1Rα protein on K562 cells gradually increased. The expression of IL-1Rα protein in 5 Gy and 10 Gy groups was significantly higher than that at 24 hours after irradiation. Conclusions Elevated IL-1 1α protein and gene expression at 24 h after irradiation may be an adaptive response to self-protection of the body and provide a material basis for early application of IL-1 1
