目的:确定藕节炭止血作用的有效部位和成分的基础上,探讨有效部位及其成分的止血作用机制。方法:大鼠灌胃给药(10.8g/kg),连续给药1周,末次用药后1 h0,.3%戊巴比妥腹腔注射麻醉,腹主动脉采血,0.109 mol/L枸橼酸钠抗凝(1:9体积)3,000 rpm离心10 min,分离血浆。测定血浆凝血酶原时间(PT),活化部分凝血活酶时间(APTT),纤维蛋白原(FIB)及D-二聚体含量,血小板聚集功能及血液流变学指标。EDTA-Na2抗凝,3000rpm离心10 min,分离血浆,放免法测定血浆TXB2和6-Keto-PGF1a。结果:三个受试药物不同程度缩短凝血酶原时间和部分活化凝血酶原时间,增加血浆纤维蛋白原的含量,降低血浆中纤维蛋白降解产物的浓度;提高血浆血栓烷B2含量,降低6-酮-前列腺素F1a含量。血小板聚集功能试验可见三个给药组不同程度提高血小板聚集率,对各受试药物进行血液流变学主要指标观察,可见血液粘度不同程度增加,红细胞电泳时间不同程度延长。结论:藕节凝血作用靶点涉及到凝血、抗凝及其血栓形成的整个过程,并由此导致血流动力学的改变,起到凝血的功效。但其有效成分表白桦脂酸的凝血作用靶点少,可能只对凝血、抗凝和血栓形成过程中某些部位起作用。
Objective: To determine the effective parts and components of hemostatic effect of lotus root charcoal, and to explore the mechanism of hemostasis of effective parts and components. Results: The three tested drugs reduced prothrombin time and partial prothrombin time, increased the content of fibrinogen and decreased the concentration of fibrin degradation products in plasma, increased the content of thromboxane B2 and decreased the content of 6- Keto-prostaglandin F1a content. Platelet aggregation test showed three groups to varying degrees of platelet aggregation rate, the main indicators of the tested drugs for the study of hemorheology, we can see the blood viscosity increased to varying degrees, the erythrocyte electrophoresis time to varying degrees. Conclusion: The target of coagulation of lotus root joints involves the whole process of coagulation, anticoagulation and thrombosis, which leads to the change of hemodynamics and coagulation effect. However, its active ingredient shows betulinic acid coagulation target less, may only coagulation, anticoagulation and thrombosis in some parts of the process.
