目的通过实验研究观察肾衰泻浊丸对腺嘌呤致肾间质纤维化大鼠骨成型蛋白-7(BMP-7)及受体(BMP-7RⅡ)的影响,探讨肾衰泻浊丸抗肾间质纤维化的机制。方法雄性Wistar成年大鼠68只,随机取8只正常大鼠作为空白对照组,其余采用腺嘌呤致肾间质纤维化大鼠模型,并随机分为模型组、尿毒清对照组、肾衰泻浊丸低剂量组、肾衰泻浊丸中剂量组和肾衰泻浊丸高剂量组。药物干预60d,观察实验大鼠治疗后肾间质纤维化情况,分别采用免疫组织化学法、Western Blot法、半定量RT-PCR检测肾组织中BMP-7及BMP-7RⅡ的表达情况。结果肾衰泻浊丸能够改善肾小管间质损伤,能够升高CRF大鼠肾组织中BMP-7及BMP-7RⅡ蛋白的表达,与对照组比较有显著性差异(P<0.01);能够上调肾组织中BMP-7mRNA及BMP-7RⅡmRNA的表达,与对照组比较有显著性差异(P<0.01),且呈剂量依存性。结论肾衰泻浊丸可能是通过增加BMP-7及BMP-7RⅡ的蛋白表达,从而减轻肾间质纤维化的程度,延缓肾间质纤维化的进展。
Objective To observe the effect of Shenshuai Xiezhu Pills on the bone protein-7 (BMP-7) and receptor (BMP-7RII) in rats with adenine-induced renal interstitial fibrosis The mechanism of interstitial fibrosis. Methods Sixty-eight male Wistar adult rats were randomly selected from 8 normal rats as the blank control group. The other rats were treated with adenine-induced renal interstitial fibrosis and randomly divided into model group, uremic control group, and renal failure. Low-dose group of turbid pills, middle-dose group of kidney-faint diarrhea pill and high-dose group of kidney-faded diarrhea pill. After 60 days of drug intervention, renal interstitial fibrosis was observed in experimental rats. Immunohistochemistry, Western Blot, and semi-quantitative RT-PCR were used to detect the expression of BMP-7 and BMP-7RII in renal tissues. Results: Shenxie Xiezhu Pill can improve renal tubulointerstitial injury, and can increase the expression of BMP-7 and BMP-7RII protein in renal tissue of CRF rats, and there is significant difference compared with the control group (P<0.01). The expression of BMP-7 mRNA and BMP-7RII mRNA in renal tissue was significantly different from that in the control group (P<0.01), and was dose-dependent. Conclusion Shenxie Xiezhuo Pill may increase the protein expression of BMP-7 and BMP-7RII, so as to reduce the degree of renal interstitial fibrosis and delay the progression of renal interstitial fibrosis.
