本文基于大型真菌茶树菇活性抗肿瘤蛋白组分Yt的研究,旨在深入理解其如何通过调节机体免疫系统来发挥生物学功能。通过H22荷瘤小鼠模型,检测瘤内注射Yt(5 mg/kg)抑制肿瘤生长活性,诱导机体产生抗体滴度,并通过再次接种肿瘤细胞检测其是否刺激机体产生免疫记忆功能,最后通过ELISA方法检测血清细胞因子IL-2和TNF-α的表达水平变化。实验发现,瘤内注射Yt可显著抑制小鼠肿瘤生长。Yt、主要蛋白组分Yp和小分子组分Ys均不会诱导机体产生大量抗体而有毒性。Yt治疗后小鼠可获得抗肿瘤免疫记忆功能,且上调血清细胞因子IL-2的表达(84.6±37.6 pg/mL,p<0.05),TNF-α与对照组相比无显著性差异。该数据表明茶树菇活性蛋白组分Yt可激活机体免疫功能,暗示了其作为肿瘤疫苗佐剂的潜力。
Based on the study of the active antitumor protein component Yt of the large fungus Agrocybe aegerita, this paper aims to gain insight into how it exerts its biological function by regulating the body’s immune system. Ht tumor-bearing mouse model was used to detect the tumor growth activity of Yt (5 mg / kg) intratumoral injection to induce the antibody titers in the body. The mice were inoculated with the tumor cells again to detect whether it stimulated the immune memory function. Finally, Methods The levels of serum cytokines IL-2 and TNF-α were measured. The experiment found that intratumoral injection of Yt can significantly inhibit tumor growth in mice. Yt, the main protein component Yp and small molecule component Ys will not induce the body to produce a large number of antibodies and toxic. After treatment with Yt, the anti-tumor immune memory function and the expression of IL-2 in serum were increased (84.6 ± 37.6 pg / mL, p <0.05). There was no significant difference between the two groups. This data suggests that Yt, an active protein component of Agrocybe aegerita, activates immune function in the body, suggesting its potential as an adjuvant to tumor vaccines.
