目的:利用基因表达谱芯片研究U937细胞在硫化砷作用前后基因表达的差异性。方法:采用急性单核细胞白血病细胞株U937,用 cy3和cy5两种不同荧光染料,通过逆转录反应将硫化砷处理前后的U937 mRNA分别标记成两种探针,并与载有一组靶基因的基因表达谱芯片进行杂交,经扫描、计算机软件分析寻找经硫化砷作用前后表达有差异的基因。结果:筛选出与细胞骨架、代谢相关酶和细胞信号转导等相关的表达有差异的基因共7条,其中1条表达上调,6条表达下调。结论:细胞骨架和细胞信号转导的改变可能参与硫化砷促进U937细胞凋亡的过程。
OBJECTIVE: To study the gene expression differences of U937 cells before and after arsenic sulfide treatment by gene expression microarray. Methods: The U937 mRNA of U937 cells treated with As 2 S 3 was labeled with two different fluorochromes cy3 and cy5 by reverse transcription reaction, respectively, and compared with U937 cells carrying a group of target genes Gene expression microarray hybridization, scanning, computer software analysis looking for the difference between before and after the arsenic sulfide gene expression. Results: Seven genes with different expression related to cytoskeleton, metabolism related enzymes and cell signal transduction were screened out, among which one was up-regulated and the other six were down-regulated. Conclusion: The changes of cytoskeleton and cell signal transduction may be involved in the process of arsenic sulfide-induced apoptosis in U937 cells.
