目的探索 TGF α对胃癌细胞周期的促进作用,以及 PTK抑制剂对这种作用的抑制效应。方法利用细胞体外培养技术,观察TGF α对胃癌SGC7901细胞的促增殖作用;细胞经G期同步化后,用流式细胞仪检测TGF α作用不同时间的细胞周期变化,用~3H-TdR掺入法检测TGFα对细胞DNA合成的影响;加PTK抑制剂tyrphostin 51处理后用流式细胞仪检测细胞凋亡。结果TGFα对SGC7901具有明显的促增殖作用,细胞生长曲线上移:TGFα作用4、6h后S期细胞和作用8、10h后G2+M期细胞均明显增加,作用5h后DNA合成也显著高于对照组。tyrphostin 51作用48h后出现了细胞凋亡。结论TGFα能够刺激胃癌SGC7901细胞生长,促进其细胞周期进程,而tyrphostin 51通过抑制TGFα受体的PTK活性,使细胞发生凋亡。
Objective To explore the promotion effect of TGF α on the cell cycle of gastric cancer and the inhibitory effect of PTK inhibitor on this effect. Methods The in vitro culture technique was used to observe the effect of TGFα on the proliferation of gastric cancer cell line SGC7901. After the cells were synchronized by G phase, the changes of cell cycle were detected by flow cytometry. Method to detect the effect of TGFα on the DNA synthesis. After treatment with PTK inhibitor tyrphostin 51, apoptosis was detected by flow cytometry. Results TGFα had a significant effect on promoting proliferation of SGC7901 cells. The cell growth curve was up-regulated: the cells in S phase at 4 and 6 hours after TGFα treatment and the cells at G2 and M phase after 8 and 10 hours were significantly increased, and the DNA synthesis was also significantly higher after 5 hours . Tyrphostin 51 appeared after 48h apoptosis. Conclusion TGFα can stimulate the growth of gastric cancer SGC7901 cells and promote its cell cycle progression. However, tyrphostin 51 can induce cell apoptosis by inhibiting the PTK activity of TGFα receptor.
