目的分析荧光定量PCR(FQ-PCR)检测血浆EB病毒(EBV)的临床应用效果。方法选择本院2015年1月-2016年12月收治的190例患者,分析其外周血单个核细胞中EBV-DNA的含量,对所有阳性患者采取血浆进行复查,分析其差异性。结果对所有患者的临床资料进行回顾性的分析,检测得知PBMC中,17例为EB阳性标本,阳性率达到8.95%,其中包括5例鼻炎,3例鼻咽癌,4例鼻息肉,3例肺炎,2例传染性单核细胞增多症;收集阳性标本,检测血浆游离EB DNA,得知16例为阳性,分析其拷贝数明显低于PBMC中的拷贝数,P<0.05。结论采用荧光定量PCR检测技术对PBMC与血浆中EBV-DNA含量进行检测的价值显著,可对确诊EBV感染相关性疾病提供参考资料,相比血浆中EBV-DNA含量,PBMC中的EBV-DNA含量明显更高。
Objective To analyze the clinical application of fluorescent quantitative PCR (FQ-PCR) in detecting plasma Epstein-Barr virus (EBV). Methods 190 patients admitted from January 2015 to December 2016 in our hospital were analyzed for EBV-DNA content in peripheral blood mononuclear cells (PBMCs). All the positive patients were examined by plasma and the differences were analyzed. Results The clinical data of all patients were retrospectively analyzed. Among PBMCs, 17 were positive for EB, and the positive rate was 8.95%, including 5 rhinitis, 3 nasopharyngeal carcinoma, 4 nasal polyps, 3 Cases of pneumonia, 2 cases of infectious mononucleosis; collected positive specimens, plasma free EB DNA was detected, 16 were positive, the copy number was significantly lower than PBMC analysis, P <0.05. Conclusion The detection of EBV-DNA in PBMC and plasma by fluorescence quantitative PCR is of great value. It can provide references for the diagnosis of EBV-related diseases. Compared with EBV-DNA content in plasma and EBV-DNA content in PBMC Obviously higher.
