观察雌二醇对血管平滑肌增殖的影响,探讨雌激素抑制动脉粥样硬化的可能机制。方法:采用去卵巢大鼠模型,使用鼠氚标记胸腺嘧啶核苷(3H-TdR)掺入技术,在体观察不同雌激素水平对大鼠胸主动脉中膜组织3H-TdR掺入量的影响。结果:切除卵巢后血管组织中膜3H-TdR掺入量极显著增加,补充大剂量E2后掺入量极显著下降,并已低于假手术组水平(Sham180.4±106.26cpm/mgOVX612.90±262.22cpm/mg,OVZX+E241.22+14.45cpm/mg,P<0.05)。结论:雌二醇对血管平滑肌增殖有明显抑制作用,这可能是其抗AS和减少绝经后冠心病的机制之一。
To observe the effect of estradiol on the proliferation of vascular smooth muscle and to explore the possible mechanism of estrogen inhibiting atherosclerosis. Methods: The effects of different estrogen levels on the incorporation of 3H-TdR into the thoracic aorta were observed in ovariectomized rats using 3H-TdR incorporation assay . RESULTS: The amount of 3H-TdR incorporation in ovarian tissue after ovariectomy was significantly increased, and the amount of 3H-TdR incorporation was significantly decreased after the addition of high-dose E2, which was significantly lower than that of sham operation group (Sham180.4 ± 106.26cpm / mgOVX612.90 ± 262.22 cpm / mg, OVZX + E241.22 + 14.45 cpm / mg, P <0.05). Conclusion: Estradiol has a significant inhibitory effect on the proliferation of vascular smooth muscle cells, which may be one of the mechanisms of anti-AS and reducing postmenopausal coronary heart disease.
