目的探讨转染Cx43(Connexin 43)人脐血源基质细胞(hUCBDCs)对人急性淋巴细胞白血病细胞SUP-B15增殖的影响。方法重组腺病毒转染以上调人脐血源基质细胞Cx43表达,建立人脐血源基质细胞和SUP-B15细胞共培养模型,选取共培养72 h为时间点收集悬浮SUP-B15细胞,采用CCK-8检测SUP-B15细胞的增殖,流式细胞仪检测细胞凋亡及细胞周期。结果人脐血源基质细胞抑制SUP-B15细胞增殖,转染Cx43后人脐血源基质细胞的抑制作用更明显,两者比较差异具有统计学意义(P<0.05);转染Cx43人脐血源基质细胞对SUP-B15细胞凋亡率为(5.51±0.51)%;明显高于未转染组[(2.26±1.41)%]和阴性对照组[(2.97±1.58)%](P<0.05)。结论转染Cx43人脐血源基质细胞具有抑制SUP-B15细胞增殖、诱导凋亡的作用。
Objective To investigate the effects of Cx43 transfected hUCBDCs on proliferation of human acute lymphoblastic leukemia cells SUP-B15. Methods Human umbilical cord blood-derived stromal cells (Cx43) were transfected with recombinant adenovirus. The co-culture model of human umbilical cord blood-derived stromal cells and SUP-B15 cells was established. The co-cultured supernatants of SUP-B15 cells were collected at 72 h. -8 was used to detect the proliferation of SUP-B15 cells. Flow cytometry was used to detect apoptosis and cell cycle. Results Human umbilical cord blood-derived stromal cells inhibited the proliferation of SUP-B15 cells. The inhibitory effect of human umbilical cord blood-derived stromal cells transfected with Cx43 was more obvious with statistical significance (P <0.05). The transfection of Cx43 human cord blood The apoptosis rate of SUP-B15 cells was (5.51 ± 0.51)%, which was significantly higher than that of untransfected cells [(2.26 ± 1.41)%] and negative control group (2.97 ± 1.58%, P <0.05 ). Conclusion Transfection of Cx43 human umbilical cord blood-derived stromal cells can inhibit the proliferation and induce apoptosis of SUP-B15 cells.
