目的研究氟对L-02细胞脂质过氧化与DNA损伤的影响,并探讨二者之间的关系。方法体外培养的L-02细胞接触不同浓度的氟化钠24 h后,检测L-02细胞脂质过氧化物(LPO)水平、还原型谷胱甘肽(GSH)含量以及DNA损伤程度。结果各氟染毒组细胞内LPO水平明显高于对照组,而GSH含量则明显低于对照组,并且二者均与氟浓度呈现明显的剂量-效应关系;各氟染毒组L-02细胞DNA损伤程度明显高于对照组。结论氟可导致L-02细胞脂质过氧化和DNA损伤,一定浓度范围内的氟所诱导的L-02细胞脂质过氧化与DNA损伤之间呈现明显的正相关关系。
Objective To investigate the effect of fluoride on lipid peroxidation and DNA damage in L-02 cells and to explore the relationship between the two. Methods L-02 cells cultured in vitro were exposed to different concentrations of sodium fluoride for 24 h. The levels of LPO, GSH and DNA damage in L-02 cells were detected. Results The levels of LPO in each fluoride-exposed group were significantly higher than those in the control group, while the content of GSH was significantly lower than that in the control group, and both showed significant dose-effect relationship with fluoride concentration. The L-02 cells DNA damage was significantly higher than the control group. Conclusion Fluoride can induce lipid peroxidation and DNA damage in L-02 cells. Fluorine-induced L-02 lipid peroxidation and DNA damage in a certain concentration range show a significant positive correlation.
