目的建立大鼠血管内成栓脑梗死模型并对其病理特征进行评价。方法使用直流电刺激颈总动脉制造血栓,通过血流引导碎栓堵塞中动脉,制造脑栓塞模型。通过测定脑血流、脑梗死体积、动物行为学等指标,评价模型特点,并观察了组织型纤维蛋白酶原激活剂(tissue plasminogen activator,t-PA)的溶栓作用。结果脑梗死大鼠的病灶区血流量降低至基准值的30%左右,并在造模24h后表现出明显的神经功能与运动功能失调,脑组织出现明显的梗死灶。使用t-PA进行溶栓治疗后可以明显改善上述病理特征。结论大鼠血管内成栓脑梗死模型适用于进行血栓性脑梗死病理研究与药物溶栓作用评价。
Objective To establish a rat model of intra-arterial embolization of cerebral infarction and evaluate its pathological features. Methods DC was used to stimulate the common carotid artery to make thrombus. The blood stream was used to guide the broken artery to plug the middle artery. The cerebral embolism model was made. The characteristics of the model were evaluated by measuring the indexes of cerebral blood flow, volume of cerebral infarction and animal behavior, and the thrombolytic effect of tissue plasminogen activator (t-PA) was observed. Results The blood flow of focal infarction area reduced to about 30% of the baseline value. After 24h of modeling, the neurological function and motor function were obviously noticed, and obvious infarction appeared in brain tissue. The use of t-PA thrombolytic therapy can significantly improve the above pathological features. Conclusion The rat model of intra-thrombotic cerebral infarction is suitable for the pathological study of thrombotic cerebral infarction and evaluation of thrombolytic effect.
