目的 观察细胞间粘附分子( I C A M1) 蛋白在大鼠脑缺血再灌注的不同时程脑组织中的表达与中性白细胞浸润程度的关系及丹参对它们的影响。方法 S D大鼠分为3 组:假手术组、对照组及丹参组。大脑中动脉缺血2 h 再灌注2 h 、12 h、24 h 、48 h 、72 h 、7 d、14 d 后,分别进行 I C A M1 免疫组织化学及组织 H E染色。结果 在脑缺血再灌早期,脑微血管内皮细胞 I C A M1 免疫反应开始逐渐增加,再灌注48 h 达到高峰,再灌注14 d 接近正常水平,同时脑缺血区中性白细胞浸润也随之增加,在时程上与 I C A M1 表达同步。丹参组,再灌注48 h 后, I C A M1 免疫阳性血管数及中性白细胞的浸润比同时间对照组明显降低。结论 脑缺血 I C A M1 的表达与中性白细胞浸润密切相关,丹参能降低 I C A M1 的表达,抑制中性白细胞的浸润。
Objective To observe the relationship between the expression of intercellular adhesion molecule (I C A M1) protein in brain tissue and the degree of neutrophil infiltration at different time points after cerebral ischemia-reperfusion in rats and the effect of salvia miltiorrhiza on them. Methods S D rats were divided into 3 groups: sham operation group, control group and Salvia group. After 2 h, 2 h, 12 h, 24 h, 48 h, 72 h, 7 d and 14 d of reperfusion of middle cerebral artery ischemia, I C A M 1 immunohistochemistry and tissue H E staining were performed respectively. Results In the early stage of cerebral ischemia and reperfusion, the immunological reaction of cerebral microvascular endothelial cells began to increase I C A M 1, 48 h after reperfusion peaked at 14 d after reperfusion close to normal levels, while cerebral ischemic neutrophil infiltration With the increase, in the schedule and I C A M 1 expression synchronization. Salvia miltiorrhiza group, 48 h after reperfusion, I C A M 1 immunoreactive blood vessels and neutrophil infiltration than the same time the control group was significantly lower. Conclusions The expression of I C A M1 in cerebral ischemia is closely related to neutrophil infiltration. Salvia miltiorrhiza can decrease the expression of I C A M1 and inhibit the infiltration of neutrophils.
