[目的]探讨大豆异黄酮对子宫雌激素受体的影响及其雌激素样作用机制。[方法]30只大鼠行双侧去卵巢手术后随机分为阴性对照组(蒸馏水)、低、中、高剂量大豆异黄酮组(0.02mg/kg、0.10mg/kg、0.50mg/kg)和阳性对照组(0.08mgkg雌二醇),连续用药3d后取子宫,采用放射性配基结合分析法和雌激素受体结合试验检测雌激素受体数量。[结果]大豆异黄酮各浓度组放射活性与阴性对照组比较,差异有统计学意义(P≤0.05)。大豆异黄酮各浓度组雌激素受体数量与阴性对照组比较,差异无统计学意义(P≥0.05)。[结论]大豆异黄酮影响雌二醇与雌激素受体结合,但不影响雌激素受体的数量,可能是通过与雌激素受体结合显示雌激素样作用。
[Objective] To investigate the effects of soy isoflavones on estrogen receptor in the uterus and its estrogen-like mechanism. [Methods] Thirty rats were randomly divided into negative control group (distilled water), low, medium and high dosage soy isoflavone group (0.02mg / kg, 0.10mg / kg, 0.50mg / kg) And the positive control group (0.08mgkg estradiol). The uterus was taken after 3d continuous administration, and the quantity of estrogen receptor was detected by radioligand binding assay and estrogen receptor binding assay. [Result] Compared with the negative control group, the radioactivity of soy isoflavones in each concentration group had significant difference (P≤0.05). Compared with the negative control group, the quantity of estrogen receptor in each concentration of soy isoflavones had no statistical significance (P> 0.05). [Conclusion] Soy isoflavones affect the binding of estradiol to estrogen receptor, but it does not affect the quantity of estrogen receptor, which may be the result of estrogen-like interaction by binding with estrogen receptor.
