目的:研究化瘀止痛方外用对骨转移癌疼痛大鼠的镇痛作用及机制。方法:采用大鼠乳腺癌MRMT-1细胞,按Medhurst方法建立骨转移癌疼痛模型。以唑来膦酸为阳性对照,观察外用药对大鼠脊髓背角神经元原癌基因c-FOS蛋白表达、神经胶质细胞胶质细胞酸性蛋白(GFAP)表达的影响。结果:相对于假手术组,各手术组脊髓后角神经元c-FOS蛋白表达明显增高,GFAP染色阳性星形胶质细胞明显增生肥大。与模型组比较,脊髓背角c-FOS表达明显降低(P<0.05),GFAP表达变化改善。结论:外用中药对骨转移癌疼痛模型有明显镇痛作用,作用机制与拮抗伤害感受器并且抑制痛觉在脊髓水平的放大有关。
Objective: To study the analgesic effect and mechanism of Huayu Zhitong Recipe on bone metastatic pain in rats. Methods: The rat breast cancer MRMT-1 cells were used to establish the pain model of bone metastases by Medhurst method. Zoledronic acid as a positive control, to observe the external use of spinal dorsal horn neurons oncogene c-FOS protein expression, glial glial cell acidic protein (GFAP) expression. Results: Compared with sham group, the expression of c-FOS protein in spinal cord posterior horn of each operation group was significantly higher than that in sham-operation group, and GFAP-positive astrocytes significantly proliferated and hypertrophied. Compared with model group, c-Fos expression in spinal dorsal horn was significantly decreased (P <0.05), and GFAP expression was improved. Conclusion: Topical Chinese herbal medicine has obvious analgesic effect on pain model of bone metastatic carcinoma, and its mechanism is related to the antagonism of nociceptors and inhibition of the enlargement of pain sensation at spinal level.
