目的:探讨槲皮素对人肝癌细胞(HepG2)生长的干预作用以及对其细胞周期基因Cyclin D2表达的影响;方法:倒置显微镜观察药物处理前后HepG2细胞的形态学变化,噻唑蓝(MTT)分析生存率,流式细胞术检测细胞周期分布,RT-PCR检测Cyclin D2表达。结果:加入50μmol/L槲皮素作用24h后,HepG2细胞逐渐变圆,附壁疏松,脱壁,增殖变慢;MTT实验结果显示,HepG2细胞生存率和槲皮素有浓度、时间依存关系,48h的IC50约为50μmol/L(15mg/L);细胞周期分析显示,有明显的G0/G1期阻滞;RT-PCR结果显示,槲皮素作用组Cyclin D2表达显著降低。结论:槲皮素能抑制HepG2细胞生长增殖,阻滞其细胞周期,并且这种作用可能是通过影响HepG2细胞Cyclin D2基因表达实现的。
Objective: To investigate the effect of quercetin on the growth of HepG2 cells and the effect of quercetin on the expression of Cyclin D2 in HepG2 cells. Methods: The morphological changes of HepG2 cells before and after drug treatment were observed by inverted microscope. The MTT assay Survival rate, cell cycle distribution were detected by flow cytometry, and Cyclin D2 expression was detected by RT-PCR. Results: After treated with 50μmol / L quercetin for 24 hours, HepG2 cells gradually became round, and the adherent wall became loose and detached. The proliferation of HepG2 cells was slowed down. The MTT results showed that the survival rate of HepG2 cells was correlated with the concentration of quercetin and the time- The IC50 of 48h was about 50μmol / L (15mg / L). Cell cycle analysis showed that G0 / G1 phase arrest was obvious. The expression of Cyclin D2 in Quercetin group was significantly decreased by RT-PCR. Conclusion: Quercetin can inhibit HepG2 cell proliferation and arrest its cell cycle, and this effect may be through affecting the expression of Cyclin D2 in HepG2 cells.
