目的探讨毛菊苣(MJJ)根和种子2种药用部位不同比例的配伍对小鼠化碳性肝损伤和免疫性肝损伤的保护作用。方法将MJJ根(root,R)和种子(seed,S)按不同(R∶S,0∶1,1∶1,1∶2,1∶3,2∶1和1∶0)质量比例进行配伍,再以8倍体积分数70%乙醇提取后获得6种配伍提取物。建立四氯化碳(CCl4)致小鼠化学性肝损伤模型和卡介苗(BCG)加脂多糖(LPS)致小鼠免疫性肝损伤模型,将小鼠随机分为空白对照组、模型组、联苯双酯组和MJJ不同比例的配伍提取物组,各组以20mL·kg-1灌胃给药。检测各组小鼠血清中天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性,计算脏器系数,观察肝组织病理性变化。结果 MJJ根-种子以质量比1∶0(R1S0)、1∶1(R1S1)和2∶1(R2S1)配伍的乙醇提取物能明显降低CCl4致小鼠急性化学性肝损伤模型中血清AST和/或ALT活性;MJJ根-种子以R1S1和R2S1配伍的乙醇提取物亦能明显降低BCG+LPS致急性免疫性肝损伤模型中血清AST和ALT活性;MJJ根-种子以R1S1和R2S1配伍的乙醇提取物还能改善模型小鼠肝细胞坏死、变性、炎性细胞浸润等病理现象。结论 MJJ根和种子不同配伍的提取物对肝损伤有不同的改善作用,根-种子以R1S1和R2S1配伍组可有效保护肝功能和缓解肝病理变化。
Objective To investigate the protective effects of different proportions of two kinds of medicinal parts of Maoji (MJJ) root and seed on carbon-induced liver injury and immune liver injury in mice. Method The MJJ root (root, R) and the seed (S) were treated at different mass ratios (R: S, 0: 1, 1: 1, 1: 2, 1: 3, 2: 1 and 1: 0) Compatibility, and then eight times the volume fraction of 70% ethanol extraction obtained six kinds of compatibility extract. To establish a model of chemical liver injury induced by carbon tetrachloride (CCl4) in mice and immunological liver injury induced by BCG plus lipopolysaccharide (LPS) in mice, the mice were randomly divided into blank control group, model group, Benzoate group and MJJ with different proportions of compatibility extract group, each group with 20mL · kg-1 intragastric administration. Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity of each group were detected, and organ coefficient was calculated to observe the pathological changes of liver tissue. Results The ethanol extracts of MJJ roots and seeds with mass ratio of 1: 0 (R1S0), 1: 1 (R1S1) and 2:1 (R2S1) significantly reduced the levels of serum AST and CCl4 in acute liver injury induced by CCl4 / Or ALT activity; MJJ root - seeds with ethanol extract of R1S1 and R2S1 compatibility can also significantly reduce serum AST and ALT activity in BCG + LPS-induced acute immune liver injury model; MJJ root - seed with R1S1 and R2S1 compatible ethanol Extracts can also improve pathological phenomena such as liver cell necrosis, degeneration and inflammatory cell infiltration in model mice. Conclusion The extracts with different compatibility of root and seed of MJJ have different effects on liver injury. Root-seed combination of R1S1 and R2S1 can effectively protect liver function and alleviate liver pathological changes.
