目的研究肝硬变大鼠肝组织内皮素-1(ET-1)的基因 mRNA 表达的变化。方法半定量逆转录多聚酶链反应(SqRT-PCR)。结果经 SqRT-PCR 测定大鼠肝组织 ET-Ⅰ基因的 mRNA(integral optical den-sity,IOD)值:各期肝硬变大鼠模型制作2周、4周、6周、8周分别为68330±1096、73836±1125、85712±1283、89215±1366,均明显高于正常对照鼠37745±1066(P<0,01),但肝硬变大鼠各组之间的差异无统计学意义(P>0.05)。结论肝硬变门静脉高压症时大鼠肝组织内 ET-1基因的 mRNA 表达明显增加。
Objective To study the changes of mRNA expression of endothelin-1 (ET-1) in cirrhotic rat liver. Methods Semi-quantitative reverse transcriptase polymerase chain reaction (SqRT-PCR). Results The integral optical den-sity (IOD) values of liver tissue were determined by SqRT-PCR. The rat model of liver cirrhosis was made at 2 weeks, 4 weeks, 6 weeks and 8 weeks respectively at 68330 ± 1096,73836 ± 1125,85712 ± 1283,89215 ± 1366, which were all significantly higher than that of the normal control group (37745 ± 1066, P <0.01), but there was no significant difference between the groups of cirrhotic rats P> 0.05). Conclusion Liver cirrhosis portal hypertension rat ET-1 gene mRNA expression was significantly increased.
