目的:通过实验验证表面黏附分子CD133是否可以作为干细胞标志物。并通过这个试验过程浅谈小细胞肺癌细胞系H446中肿瘤干细胞的分选与鉴定。方法:在分选H446细胞(MACS方法)过程中得到CD133阳性(CD133+)细胞与CD133阴性(CD133-)细胞。对这两种细胞进行集落形成、自我更新、增殖分化、侵袭性、耐药性和成瘤性方面等项目的鉴定。结果:各种办法鉴定,发现两种细胞不存在差异。结论:分选小细胞肺癌H446细胞系中肿瘤干细胞方法中不能以表面黏附分子CD133为的标志物。
Objective: To verify whether CD133 can be used as a stem cell marker by experiments. And through this test process on the small cell lung cancer cell line H446 cancer stem cell sorting and identification. Methods: CD133 positive (CD133 +) cells and CD133 negative (CD133-) cells were obtained during the sorting of H446 cells (MACS method). The two cells were identified for colony formation, self-renewal, proliferation and differentiation, invasiveness, drug resistance and tumorigenicity. Results: A variety of methods identified and found no difference between the two cells. CONCLUSION: The method of sorting tumor stem cells in H446 cell line of small cell lung cancer can not use the surface adhesion molecule CD133 as a marker.
