目的探讨大肠癌组织中survivin的表达特征及其与血管生成的关系。方法采用免疫组化S-P法,检测64例大肠癌组织中survivin蛋白的表达情况,并选取CD34标记血管内皮细胞测定MVD。其中结肠癌34例,直肠癌30例,高-中分化腺癌42例,低分化腺癌22例,Dukes分期A-B期36例,C-D期28例,有淋巴结转移者37例,无淋巴结转移者27例。结果 64例大肠癌组织中survivin蛋白的阳性率为50%;Dukes分期C-D期和有淋巴结转移的大肠癌组织survivin的阳性率明显高于Dukes分期A-B期和无淋巴结转移的大肠癌组织,差异均有显著性意义(P<0.05);survivin表达阳性组MVD明显高于survivin表达阴性组(P<0.01)。结论 survivin的表达与大肠癌的发展,生物学行为和预后可能有关,可作为重要的生物学标志。
Objective To investigate the expression of survivin in colorectal carcinoma and its relationship with angiogenesis. Methods Immunohistochemical S-P method was used to detect the expression of survivin protein in 64 cases of colorectal carcinoma. CD34-labeled vascular endothelial cells were used to detect MVD. 34 cases of colon cancer, 30 cases of rectal cancer, 42 cases of high-moderately differentiated adenocarcinoma, 22 cases of poorly differentiated adenocarcinoma, 36 cases of Dukes stage AB, 28 cases of CD stage, 37 cases of lymph node metastasis, no lymph node metastasis 27 cases. Results The positive rate of survivin protein in 64 colorectal cancer tissues was 50%. The positive rates of survivin protein in Dukes staging CDC and lymph node metastasis were significantly higher than those in Dukes staging ABC and non-lymph node metastasis (P <0.05). The MVD in survivin positive group was significantly higher than that in survivin negative group (P <0.01). Conclusion The expression of survivin may be related to the development of colorectal cancer, biological behavior and prognosis, which may serve as an important biological marker.
