用RTPCR方法扩增并经克隆得胶质细胞源神经营养因子(glialcellinederivedneurotrophicfactor,GDNF)全长基因。实验表明,此基因的表达产物对培养的多巴胺神经元有营养和保护作用。将gdnf基因克隆入逆转录病毒载体pLNCX中,PA317包装后,感染大鼠成肌细胞株L6TG,经G418筛选得到整合并表达gdnf基因的细胞株L6TG/gdnf。此细胞株将用于帕金森病基因治疗研究。
The full length gene of glialcellinederived neurotrophic factor (GDNF) was amplified and cloned by RTPCR. Experiments show that the expression product of this gene has a nutrition and protective effect on cultured dopamine neurons. The gdnf gene was cloned into retroviral vector pLNCX, PA317 packaging, infected rat myoblast L-6TG, G418 screening and integration and expression of gdnf gene cell line L 6TG / gdnf. This cell line will be used to study Parkinson’s disease gene therapy.
