目的 研究人胰腺癌细胞中nm23 mRNA的表达与其转移潜能的关系。 方法 在建立不同转移潜能的人胰腺癌JF305 单克隆细胞亚系的基础上,应用定量逆转录多聚酶链式反应技术(RTPCR)检测不同转移潜能的人胰腺癌JF305 单克隆细胞亚系细胞中nm23 mRNA 的表达。 结果 nm23H1 基因mRNA在低转移潜能的JF305C细胞系中的表达(1-90 ±0-09)明显高于其在高转移潜能的JF305A 细胞系中的表达(1-63±0-10,P< 0-01);nm23H2 基因mRNA的表达,在高、低转移潜能的细胞亚系间差异无显著意义(0-79 ±0-10 ,0-77±0-06,P>0-05)。 结论 nm23 基因,主要是nm23H1 基因,在人胰腺癌中起转移抑制作用
Objective To study the relationship between the expression of nm23 mRNA and its metastatic potential in human pancreatic cancer cells. METHODS: Based on the establishment of human pancreatic cancer JF305 monoclonal cell sublines with different metastatic potential, Quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to detect human pancreatic cancer JF305 monoclonal cell sublines with different metastatic potentials. Nm23 mRNA expression. Results The expression of nm23-H1 mRNA in the JF305C cell line with low metastatic potential (1-90 ± 0-09) was significantly higher than that in the high metastatic potential JF305A cell line (1-63±0-10, P<0-01); nm23H2 mRNA expression was not significantly different between high and low metastatic potential subpopulations (0-79±0-10, 0-77±0-06, P>0 -05). Conclusion The nm23 gene, mainly the nm23H1 gene, plays a role in metastasis inhibition in human pancreatic cancer.
