目的:应用荧光标记的信使核糖核酸(messengerribonucleicacid,mRNA)差异显示技术分离子宫肌瘤及正常子宫肌组织相关的差异基因片段。方法:用4种锚定引物和3种随机引物,共12种组合,结合GenomyxLRTMDNA差异显示系统及GenomyxSCTM荧光图像扫描系统,以及配套的FluoroDDandHI-EROGLYPHmRNAProfile试剂盒,采用荧光标记的mRNA差异显示技术进行子宫肌瘤及正常子宫肌组织相关差异基因片段的分离。结果:从子宫肌瘤及正常子宫肌组织间分离出相关的差异基因片段27条,差异片段中主要表现为条带密度的强弱差别。结论:子宫肌瘤的产生可能与某些基因表达的差异有关,而差异基因的序列及功能有待进一步研究。
OBJECTIVE: To screen differentially expressed genes related to uterine fibroids and normal uterine muscles by fluorescently labeled messenger ribonucleic acid (mRNA) differential display technique. Methods: Four kinds of anchored primers and three kinds of random primers were used in combination with GenomyxLRTM DNA differential display system and GenomyxSCTM fluorescence image scanning system, and matched FluoroDDandHI-EROGLYPH mRNAProfile kit. Fluorescently labeled mRNA differential display technique was used to detect the uterus Isolation of Differentially Expressed Gene Fragments from Fibroids and Normal Uterine Muscles. Results: Twenty-seven differential gene fragments were isolated from uterine leiomyoma and normal uterine myometrium. The differences among the different fragments were mainly the differences in the intensity of the bands. Conclusion: The production of uterine fibroids may be related to the differences in the expression of some genes, but the sequence and function of the genes to be further studied.
