目的观察双黄糖脂宁片去蒽醌提取后对自发性2型糖尿病(GK)大鼠糖脂代谢紊乱的影响。方法选用10只健康雄性Wistar大鼠作为正常组,将36只GK大鼠根据空腹血糖(FPG)并参考糖负荷2小时血糖(2hPG)分为模型组、中药低剂量组、中药高剂量组和二甲双胍组,每组9只,予饮用水或药物灌胃4周;4周时测FPG、2hPG、随机血糖(RPG),检测血脂4项[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)]、糖化血红蛋白(HbAlc)和肝脏三酰甘油(L-TG),检测血清胰岛素(Ins)并计算胰岛素敏感指数(ISI)。结果与正常组比较,模型组大鼠FPG、2hPG、RPG、HbAlc、血清TC、LDL-C、L-TG均显著升高,Ins、ISI显著降低(P<0.01)。与模型组比较,各用药组均能显著提升ISI水平、降低HbAlc水平(P<0.05);中药各剂量组RPG、L-TG水平均显著降低(P<0.01);中药低剂量组TG水平显著降低(P<0.05);中药高剂量组Ins、TC水平显著降低(P<0.01);二甲双胍组RPG、TG、L-TG、ISI亦显著降低(P<0.05)。结论去蒽醌双黄糖脂宁片能显著改善GK大鼠糖脂代谢紊乱,具有良好的开发前景。
Objective To observe the effect of Shuanghuangzhizhengning tablet on the disturbance of glucose and lipid metabolism in spontaneously type 2 diabetic rats after its removal of anthraquinone. Methods Ten healthy male Wistar rats were selected as the normal group. 36 GK rats were divided into model group, fasting blood glucose (FPG) group and high glucose group (2hPG) Metformin group, with 9 rats in each group. The animals were given gavage with drinking water or drugs for 4 weeks. FPG, 2hPG and RPG were measured at 4 weeks, and the levels of total cholesterol (TC), triglyceride (TG) (LDL-C) and high-density lipoprotein cholesterol (HDL-C)], HbAlc and L-TG, serum insulin was measured and Insulin sensitivity index (ISI). Results Compared with normal group, FPG, 2hPG, RPG, HbAlc, TC, LDL-C and L-TG in model group were significantly increased, while Ins and ISI were significantly decreased (P <0.01). Compared with the model group, the levels of ISI and the level of HbA1c in each group were significantly increased (P <0.05); the levels of RPG and L-TG in each dose group were significantly decreased (P <0.01) (P <0.05). The levels of Ins and TC in high-dose group were significantly decreased (P <0.01). The levels of RPG, TG, L-TG and ISI in metformin group were also significantly decreased (P <0.05). Conclusion Anthraquinone Shuanghuanglianning tablet can significantly improve the disorder of glucose and lipid metabolism in GK rats and has good development prospect.
