为了深入了解小麦育种亲本SW3243的遗传基础,利用多重PCR引物对SW3243及其他47个材料进行1BL/1RS易位系检测,同时对SW3243进行Giemsa-C带分析,并利用5对定位于黑麦染色体1RS上的特异PCR引物对1BL/1RS易位系进行了多样性分析。多重PCR分析结果表明,包括SW3243在内共23个材料含有1BL/1RS易位。细胞学研究结果证实,SW3243含有1对1BL/1RS易位染色体;筛选到定位于黑麦1RS上的3对SSR引物(Xmwg913、Xmwg2062a、SCM9)可用于区别不同的1BL/1RS易位染色体;用筛选到的3对特异PCR引物对SW3243及其他22个1BL/1RS品种(系)进行分析,结果表明SW3243所含1BL/1RS易位染色体与SW22514、西科麦2号、山前麦、川麦32、川麦35的1 BL/1 RS易位染色体不同。
In order to further understand the genetic basis of SW3243 breeding parents, multiplex PCR primers were used to detect the 1BL / 1RS translocation line in SW3243 and 47 other materials. Meanwhile, G329 gene of SW3243 was analyzed by Giemsa-C banding. 1RS specific primers on the 1BL / 1RS translocation lines were analyzed for diversity. Multiplex PCR analysis showed that a total of 23 materials including SW3243 contained 1BL / 1RS translocation. Cytological results confirmed that SW3243 contains 1 pair of 1BL / 1RS translocation chromosomes. Three pairs of SSR primers (Xmwg913, Xmwg2062a, SCM9) screened to locate on rye 1RS can be used to distinguish different 1BL / 1RS translocations; Three pairs of specific PCR primers were used to analyze SW3243 and other 22 strains of 1BL / 1RS. The results showed that 1BL / 1RS translocation chromosome contained in SW3243 was associated with SW22514, Sikomai 2, Pimara, Chuanmai 32, Chuanmai 35 1 BL / 1 RS translocation chromosomes different.
