目的观察蛛网膜下腔注射Plv-UbC-HPPE/HEK293细胞对骨癌痛大鼠行为学以及脊髓背角谷氨酸/天冬氨酸转运体(GLAST)表达的影响。方法 60只健康雌性SD大鼠,随机分为4组(n=15),假骨癌空白对照组(A)、骨癌模型组(B)、骨癌模型鞘内注射UbC-GFP-LV/HEK293对照组(C)、骨癌模型鞘内注射Plv-UbC-HPPE/HEK293细胞治疗组(D)。造模术前1d,术后1、5、10、15、20、21、23、25d测定各组大鼠后肢缩足潜伏期(Paw withdrawal latency,PWL)。C组和D组分别于造模术后第20d蛛网膜下腔注射UbC-GFP-LV/HEK293细胞和Plv-UbC-HPPE/HEK293细胞。造模术后25d取各组大鼠L4-6脊髓,以免疫组化方法检测脊髓背角GLAST的表达。结果 B、C组大鼠与对照组(A)相比,大鼠PWL逐渐降低(P<0.05),GLAST表达降低(P<0.05);D组大鼠鞘内注射Plv-UbC-HPPE/HEK293细胞后PWL升高(P<0.05),GLAST蛋白表达明显升高(P<0.05)。结论 Plv-UbC-HPPE/HEK293可以增加大鼠脊髓背角GLAST蛋白表达,可能是治疗骨癌疼痛的机制之一。
Objective To investigate the effects of subarachnoid injection of Plv-UbC-HPPE / HEK293 cells on the behavior and the glutamate / aspartate transporter (GLAST) expression in spinal pain rats. Methods Sixty healthy female SD rats were randomly divided into 4 groups (n = 15), blank control group (A), bone cancer model group (B), intrathecal injection of UbC-GFP-LV / HEK293 control group (C), bone cancer model intrathecal Plv-UbC-HPPE / HEK293 cell treatment group (D). Paw withdrawal latency (PWL) was measured at 1 day before operation and at 1, 5, 10, 15, 20, 21, 23 and 25 days after operation. Groups C and D were injected with UbC-GFP-LV / HEK293 cells and Plv-UbC-HPPE / HEK293 cells subcutaneously 20 days after modeling. Twenty-five days after modeling, L4-6 spinal cord of each group was taken and the expression of GLAST in spinal dorsal horn was detected by immunohistochemistry. Results Compared with the control group (A), the PWL of rats in group B and C were decreased (P <0.05) and the expression of GLAST was decreased (P <0.05). In group D, the levels of Plv-UbC-HPPE / HEK293 After the cells, PWL increased (P <0.05), GLAST protein expression increased significantly (P <0.05). Conclusion Plv-UbC-HPPE / HEK293 can increase GLAST protein expression in the dorsal horn of the spinal cord in rats, which may be one of the mechanisms for the treatment of bone cancer pain.
