目的用温阳解郁三种不同组方对母婴分离/社会击败应激抑郁症小鼠模型进行干预治疗,探讨其作用机理。方法选用从出生起连续10 d、每天3h母婴分离的12~20周成年C57BL/6J小鼠遭受社会击败应激塑造抑郁症模型。分别以逍遥散、二仙汤和温阳解郁复方(XSF)进行6周的干预,放射免疫测定正常状态下和DEX/CRH试验血清皮质酮水平,Western blot检测海马区糖皮质激素受体(glucocorticoid reccptor,GR)含量。结果正常状态下模型组的皮质酮水平并无明显变化(P>0.05);Dex/CRH试验中,模型组的皮质酮水平异常抬高,XSF组、氟西汀组明显降低。MD小鼠海马的GR明显减少,而二仙汤、XSF可上调这种变化。结论温阳解郁复方对抑郁症有明显治疗作用,可能是温阳和解郁互相配合的结果。
Objective To investigate the mechanism of action of WenYangXieYu three different prescriptions on the mouse model of maternal and child separation / social defeat stress depression. Methods Adult C57BL / 6J mice aged 12-20 weeks separated from their mothers and their infants for 3 consecutive days from birth were challenged with social defeat stress to create depression model. The interventions of Xiaoyao San, Erxian Decoction and Wenyang Jieyu Compound (XSF) for 6 weeks were performed respectively. Serum corticosterone level in normal condition and DEX / CRH test were measured by radioimmunoassay. Western blot was used to detect glucocorticoid receptor glucocorticoid reccptor, GR) content. Results There was no significant difference in corticosterone level between model group and model group (P> 0.05). In Dex / CRH model, the level of corticosterone in model group was abnormally elevated, while it was significantly decreased in XSF group and fluoxetine group. GR in hippocampus of MD mice decreased significantly, while ErShu Decoction and XSF could up-regulate this change. Conclusions Wenyang Jieyu Compound has a significant therapeutic effect on depression, which may be the result of the combination of Wenyang and Xieyu.
