用琼脂糖双扩散法测得抗人精子单克隆抗体HS11、HS63和抗小鼠精子单抗MS204,MS207属IgG_1,抗小鼠精子单抗MS79、MS212属IgM。将上述单抗通过腹腔注射被动免疫正常CD-1雄性小鼠,然后收集受鼠血清及其睾丸、附睾各段的精子作免疫荧光染色。发现所有抗精子单抗均能进入血循环,但被血-睾屏障所阻挡。HS11、HS63、MS204和MS207能进入附睾管腔,与相应的精子抗原发生免疫反应,但染色百分率较低。MS79、MS212与附睾各段精子均无染色反应。提示血-附睾屏障能阻止IgM,但不排斥IgG_1。
Anti-human sperm monoclonal antibody HS11, HS63 and anti-mouse sperm monoclonal antibody MS204, MS207 IgG1, anti-mouse sperm monoclonal antibody MS79, MS212 IgM were measured by agarose double diffusion method. The above McAb was passively immunized normal CD-1 male mice by intraperitoneal injection, and then the sperm from the serum of the rats and the testis and epididymis were collected for immunofluorescence staining. All anti-sperm mAb was found to enter the bloodstream, but was blocked by the blood-testis barrier. HS11, HS63, MS204 and MS207 can enter the lumen of the epididymis and react with the corresponding sperm antigens, but the staining percentage is low. MS79, MS212 and epididymal sperm were no staining reaction. Prompt blood - epididymis barrier can prevent IgM, but does not exclude IgG_1.
