目的 :探讨核转录因子 κB(NF κB)及肿瘤坏死因子 α(TNF α)在大鼠视网膜缺血再灌注损伤中的表达及吡咯醛二硫氨基甲酸 (pyrrolidinedithiocarbamate ,PDTC)对其表达的影响作用。方法 :建立大鼠视网膜缺血再灌注模型 ,6 0只SD大鼠随机分为缺血再灌注组和缺血再灌注 +PDTC组。每组按再灌注后不同时间段分为 1、6、12、2 4、4 8、72小时组 ,每组 5只 ,以原位杂交法检测视网膜中NF κB和TNF α的表达 ,每只大鼠处死前行视网膜电图 (ERG)检测。结果 :缺血再灌注组在 6小时开始检测到NF κB和TNF α的表达 ,在 2 4小时表达最强 ,以后逐渐减弱。缺血再灌注 +PDTC组在再灌注 6小时未能检测到NF κB和TNF α的表达 ,在第 12小时有NF κB和TNF α的表达 ,2 4小时表达最强 ,但低于缺血再灌注组 (P <0 0 5 )。缺血再灌注组在再灌注 6小时后各期ERG波幅明显低于缺血再灌注 +PDTC组 (P <0 0 5 )。结论 :NF κB及其诱导的TNF α在视网膜缺血再灌注损伤中发挥重要作用 ,PDTC可能通过抑制NF κB的活性减轻视网膜缺血再灌注损伤
Objective: To investigate the expression of nuclear factor kappa B (NF κB) and tumor necrosis factor α (TNF α) in the rat retina after ischemia-reperfusion injury and the effect of pyrrolidineithiocarbamate (PDTC) on its expression . Methods: The rat model of retinal ischemia-reperfusion was established. Sixty SD rats were randomly divided into ischemia-reperfusion group and ischemia-reperfusion + PDTC group. Each group according to different time after reperfusion were divided into 1,6,12,24,48,72 hours group, 5 rats in each group, in situ hybridization detection of retinal NFκB and TNFα expression, each Electrocardiogram (ERG) detection was performed before the rats were sacrificed. Results: The expressions of NF-κB and TNF-α were detected in the ischemic reperfusion group at 6 hours, and reached the peak at 24 hours and then gradually weakened. The expressions of NF-κB and TNF-α were not detected in IR + PDTC group at 6 hours after reperfusion. The expressions of NF-κB and TNF-α were detected at 12 hours, and reached the peak at 24 hours, but lower than that of ischemia Perfusion group (P <0 05). The amplitude of ERG in each group after ischemia-reperfusion was significantly lower than that in ischemia-reperfusion + PDTC group (P <0.05) at 6 hours after reperfusion. CONCLUSION: NFκB and its induced TNFα play an important role in retinal ischemia-reperfusion injury. PDTC may reduce the damage of retinal ischemia-reperfusion injury by inhibiting the activity of NFκB
