目的:探讨在体外缺氧环境下缺氧诱导因子1α(HIF-1α)对肝细胞生长因子(HGF)的影响。方法:选择人前列腺癌LNCaP细胞为研究对象,对其进行低氧培养,以常氧培养为对照,在HIF-1α抑制剂YC-1作用后,检测培养液上清HGF的水平。在低氧培养条件下,应用YC-1抑制HIF-1α表达后,观察低氧对HGF水平的影响,并进行比较。结果:人前列腺癌LNCaP细胞在低氧条件下HGF的表达水平为(465.37±59.61)ρg/mL,显著高于常氧培养(240.52±47.52)ρg/mL,P<0.01。在低氧培养条件下,应用YC-1抑制HGF表达后,HGF表达水平为(438.62±61.93)ρg/mL,未见明显变化,P>0.05。结论:低氧条件下,人前列腺癌细胞LNCaP细胞HGF表达上调可能不受HIF-1α的影响。因此,在低氧条件下,可能存在其他调节通路影响HGF表达。
Objective: To investigate the effect of hypoxia inducible factor 1α (HIF-1α) on hepatocyte growth factor (HGF) in vitro under anoxic conditions. Methods: Human prostate cancer cell line LNCaP was selected as the research object. Hypoxia was used to culture HNCs and the normoxic culture was used as the control. After HIF-1α inhibitor YC-1 was administered, the level of HGF in the culture supernatant was detected. Under the condition of hypoxia culture, the effect of hypoxia on the level of HGF was observed and compared after YC-1 was used to inhibit the expression of HIF-1α. Results: The expression of HGF in human prostate cancer LNCaP cells was (465.37 ± 59.61) ρg / mL under hypoxia, which was significantly higher than that of normoxic culture (240.52 ± 47.52 ρg / mL, P <0.01). Under hypoxic conditions, the expression level of HGF was inhibited by YC-1 (438.62 ± 61.93 ρg / mL), with no significant change (P> 0.05). CONCLUSION: Hypoxia may up-regulate the expression of HGF in human prostate cancer cell line LNCaP cells without HIF-1α. Therefore, under hypoxic conditions, there may be other regulatory pathways affecting HGF expression.
