目的:观察程氏蠲痹汤加减对佐剂关节炎大鼠一般形态学改变、血清肿瘤坏死因子α(TNF-α)的影响以及病理变化。方法:24只雄性SD大鼠随机分成4组:正常对照组、模型组、程氏蠲痹汤加减(JBT)组、雷公藤多苷片(TGT)组。弗氏完全佐剂造模,同时冷水吹风复制风寒湿型痹病,两者结合复制病+证模型。观测大鼠一般形态学改变,并采用酶联免疫吸附试验(ELISA)法检测各组大鼠血清TNF-α水平,制作关节滑膜的石蜡蜡块,病理切片,并进行HE染色处理。结果:JBT组大鼠足爪肿胀度和关节炎指数明显减轻,与模型组相比有非常显著性差异(P<0.01)。模型组大鼠血清TNF-α含量明显增加,与正常组相比有非常显著性差异(P<0.01)。JBT组大鼠血清TNF-α含量明显降低,与模型组相比有非常显著性差异(P<0.01)。HE染色切片显示变化较大。结论:TNF-α在RA滑膜组织炎症形成和发展中发挥着重要作用,程氏蠲痹汤加减能下调血清TNF-α水平,药效明显。
Objective: To observe the effects of Chengshi Tongbi decoction on the general morphological changes, serum tumor necrosis factor α (TNF-α) and pathological changes in adjuvant arthritis rats. Methods: Twenty-four male Sprague-Dawley rats were randomly divided into 4 groups: normal control group, model group, JBT group and TGT group. Freund’s complete adjuvant model, while cold water copy of rheumatoid paralysis, both combined with replication disease + syndrome model. The morphological changes of rats were observed. The levels of TNF-α in serum of rats in each group were measured by enzyme linked immunosorbent assay (ELISA), paraffin wax blocks and pathological sections of synovial membrane were made and HE staining was performed. Results: The degree of paw swelling and the arthritis index of the JBT group were significantly reduced compared with the model group (P <0.01). The content of TNF-α in model group increased significantly, which was significantly different from the normal group (P <0.01). Compared with model group, the level of TNF-α in serum of JBT group was significantly decreased (P <0.01). HE stained sections showed a large change. CONCLUSION: TNF-α plays an important role in the formation and development of RA synovial tissue inflammation. The effect of Chengbi Huibi Decoction on the level of serum TNF-α can be reduced by addition and subtraction.
