目的:研究肢体缺血预处理(limb ischemic preconditioning,LIP)对成年大鼠脑梗死后海马区神经干细胞的激活增殖作用。方法:建立大鼠脑梗死模型,观察LIP后不同时段的脑梗死大鼠海马神经干细胞数量的变化。48只大鼠随机分为4组:(1)对照组(n=12),假预处理+脑梗死;(2)LIP+脑梗死,根据LIP与脑梗死间隔时间不同,分为12、24、48h组。所有大鼠在脑梗死后24h,断头取脑组织切片,免疫组化染色,高倍镜下观察梗死侧海马Nestin阳性细胞数,随机3个视野,取其平均数。结果:对照组Nestin阳性细胞数为(27.33±3.38);LIP12、24、48h组分别为(32.94±6.12)、(50.61±6.11)和(52.17±6.80),与对照组比较,差异均有统计学意义(P<0.05),表明LIP后使脑梗死大鼠海马的神经干细胞数量增加,且在12~48h内持续增加。结论:LIP后12~48h可以激活脑梗死大鼠海马的神经干细胞增殖,可能对脑梗死后机体自我修复有潜在的治疗作用。
Objective: To investigate the effect of limb ischemic preconditioning (LIP) on the proliferation of neural stem cells in hippocampus of adult rats after cerebral infarction. Methods: The rat model of cerebral infarction was established and the changes of the number of hippocampus neural stem cells in different stages after LIP were observed. 48 rats were randomly divided into 4 groups: (1) control group (n = 12), fake preconditioning + cerebral infarction; (2) LIP + cerebral infarction, according to LIP and cerebral infarction interval is divided into 12,24, 48h group. All rats were sacrificed 24h after brain infarction, brain sections were taken out, immunohistochemical staining, high magnification at the infarction side of the hippocampal Nestin positive cells, random 3 fields, whichever is the average. Results: The number of Nestin positive cells in the control group was (27.33 ± 3.38), that in the control group was (32.94 ± 6.12), (50.61 ± 6.11) and (52.17 ± 6.80), respectively (P <0.05), indicating that after LIP, the number of neural stem cells in the hippocampus of cerebral infarction rats increased and continued to increase within 12 ~ 48h. CONCLUSION: After 12 ~ 48h of LIP, the proliferation of neural stem cells in the hippocampus of cerebral infarction rats may be activated, which may have a potential therapeutic effect on the self-repair of the body after cerebral infarction.
