目的探讨热疗增强顺铂(DDP)对子宫颈癌细胞的作用。方法以体外培养的子宫颈癌Hela细胞株为研究对象,采用组内分组设计方法,以恒温水浴热疗,藉MTT法观测不同热疗温度、时间、序贯对DDP化疗增效作用。结果(1)Hela细胞在37℃下与0.1mg·L-1DDP作用24h,细胞毒指数(CI)为0.08,辅以43℃热疗30min,CI值提高8.5倍;而DDP浓度提高1000倍时,CI值仅提高10倍。43℃热疗组与41℃以下热疗组间的CI值差异有显著性(P<0.01)。(2)热疗时间15min以上组与7.5min组之间的CI值差异有显著性(P<0.01),而15min以上各组间CI值差异无显著性(P>0.05)。(3)热疗与DDP同时应用者细胞毒性作用最强。结论热疗增强DDP对Hela细胞的毒性作用,以43℃15min以上热疗与DDP同时应用效果最佳。
Objective To investigate the effect of hyperthermia and cisplatin (DDP) on cervical cancer cells. Methods In vitro cultured cervical cancer Hela cell line was used as research object. Intragroup grouping design method was used to heat therapy with constant temperature water bath. MTT method was used to observe the synergistic effect of different hyperthermia temperature, time and sequential chemotherapy on DDP. Results (1) Hela cells were treated with 0.1 mg·L-1 DDP at 37°C for 24 h, the cytotoxicity index (CI) was 0.08, and the hyperthermia at 43°C for 30 min increased the CI value by 8.5 times. When the concentration is increased by 1000 times, the CI value only increases by 10 times. There was significant difference in CI values between the 43°C hyperthermia group and the hyperthermia group below 41°C (P<0.01). (2) There was a significant difference in the CI value between the group with 7.5 min and the group with hyperthermia for 15 min (P<0.01), but there was no significant difference between the groups in the CI value between 15 min and above (P>0.05). . (3) Patients with hyperthermia combined with DDP had the strongest cytotoxicity. Conclusion Hyperthermia enhances the toxic effect of DDP on Hela cells. The effect of hyperthermia with DDP at 43°C for 15 min is best.
