目的建立高效液相色谱法测定大花罗布麻叶中异槲皮苷含量的方法。方法采用高效液相色谱法对样品中异槲皮苷进行定量分析,以十八烷基硅烷键合硅胶为填充剂;甲醇-乙腈-0.3%磷酸(11∶10∶79)为流动相,柱温为35℃,检测波长为354 nm。结果异槲皮苷进样量在浓度为0.020 2~0.202 0 mg/m L(r=0.999 9)范围内与其峰面积呈良好线性关系。平均回收率为104.45%(n=6),RSD为0.45%。10批不同产地样品中异槲皮苷的含量为0.320%~0.392%,平均含量为0.37%。结论 HPLC色谱法对大花罗布麻叶中异槲皮苷进行测定,方法简便,精确,重现性好,为有效制订质量标准提供科学依据。
Objective To establish a HPLC method for the determination of isoquercitrin in Apocynum leaves. Methods The content of isoquercitrin in samples was determined by HPLC. The octadecylsilane bonded silica gel was used as the filler. Methanol-acetonitrile-0.3% phosphoric acid (11:10:79) was used as the mobile phase. The temperature is 35 ℃, the detection wavelength is 354 nm. Results The isoquercitrin injection showed a good linear relationship with the peak area in the range of 0.020 2 ~ 0.202 0 mg / m L (r = 0.999 9). The average recovery was 104.45% (n = 6) with a RSD of 0.45%. The content of isoquercitrin in 10 batches of different producing areas was 0.320% -0.392% with the average content of 0.37%. Conclusion The HPLC method for the determination of isoquercitrin in Apocynum leaves is simple, accurate, reproducible and provides a scientific basis for the effective formulation of quality standards.
