目的研究野黄芪甙原对实验性糖尿病大鼠肾脏蛋白激酶C(PKC)活性、肾功能及肾脏结构的影响。方法大鼠随机分为正常对照组(A组)、糖尿病组(B组)、野黄芪甙原治疗组(C组),分别于4、6周应用酶联免疫法测定肾脏PKC活性;同时测定尿蛋白、Ccr、Scr及肾脏肥大指数,以光镜及电镜观察肾脏组织。结果C组尿蛋白排泄率、Ccr、肾脏肥大指数及细胞膜PKC活性均低于B组(P均<0.05);光镜、电镜病理改变C组较B组有所改善。结论野黄芪甙原对糖尿病大鼠肾脏病变有部分保护作用,其机制可能部分通过下调肾脏PKC活性。
Objective To study the effect of wild astragalosides on renal protein kinase C (PKC) activity, renal function and renal structure in experimental diabetic rats. Methods The rats were randomly divided into normal control group (group A), diabetic group (group B) and wild astragaloside treatment group (group C). PKC activity was measured at 4 and 6 weeks by enzyme-linked immunosorbent assay Urine protein, Ccr, Scr and renal hypertrophy index were observed by light microscope and electron microscope. Results The urinary protein excretion rate, Ccr, renal hypertrophy index and cell membrane PKC activity in group C were lower than those in group B (all P <0.05). The pathological changes in group C were better than those in group B by light microscope and electron microscope. CONCLUSIONS: Astragaloside has some protective effects on the pathological changes of kidney in diabetic rats, and its mechanism may be partly through the down-regulation of PKC activity in kidney.
